Modafinil, sold under the brand name Provigil among others, is a medication to treat sleepiness due to narcolepsy, shift work sleep disorder, and obstructive sleep apnea. While it has seen off-label use as a purported cognitive enhancer to improve wakefulness in animal and human studies, the research on its effectiveness for this use is not conclusive. Side Effect Of Modafinil

Modafinil is generally well-tolerated, with possible side effects including headache, trouble sleeping, and nausea. Rare serious dermatological reactions may occur in people with genetic predispositions and more commonly in pediatric patients, and, discontinuation is recommended at the earliest signs of rash or similar reactions. Serious rare side effects may include allergic reactions such as anaphylaxis and Stevens–Johnson syndrome, as well as hallucinations in people with a history of psychosis. It is unclear if use during pregnancy is safe. The amount of medication used may need to be adjusted in those with kidney or liver problems.  It is not recommended in those with arrhythmia, significant hypertension, or left ventricular hypertrophy. Modafinil appears to work by acting on dopamine and modulating the areas of the brain involved with the sleep cycle.

Modafinil Usage

  • Take modafinil exactly as prescribed by your doctor. Your doctor will prescribe the dose of modafinil that is right for you. Do not change your dose of modafinil without talking to your doctor.
  • Your doctor will tell you the right time of day to take modafinil.
  • People with narcolepsy or obstructive sleep apnea (OSA) usually take modafinil one time each day in the morning.
  • People with shift work disorder (SWD) usually take modafinil about 1 hour before their work shift.
  • Do not change the time of day you take modafinil unless you have talked to your doctor. If you take modafinil too close to your bedtime, you may find it harder to go to sleep.
  • You can take modafinil with or without food.

Overdose

In mice and rats, the median lethal dose (LD50) of modafinil is approximately or slightly greater than 1250 mg/kg. Oral LD50 values reported for rats range from 1000 to 3400 mg/kg. Intravenous LD50 for dogs is 300 mg/kg. Clinical trials on humans involving taking up to 1200 mg/day for 7–21 days and known incidents of acute one-time overdoses up to 4500 mg did not appear to cause life-threatening effects, although a number of adverse experiences were observed, including excitation or agitation, insomnia, anxiety, irritability, aggressiveness, confusion, nervousness, tremor, palpitations, sleep disturbances, nausea, and diarrhea. As of 2004, the FDA is not aware of any fatal overdoses involving modafinil alone (as opposed to multiple drugs including modafinil).

Missed Dose

If you miss a dose, take it as soon as you remember. If it is near the time of the next dose, skip the missed dose. Take your next dose at a regular time. Do not double the dose to catch up. Do not take missed doses close to bedtime because doing so may make it harder to go to sleep.

Store at room temperature away from light and moisture. Do not store in the bathroom. Keep all medications away from children and pets.
